The essential oils of three wild-growing Thymus species, collected from west of Iran during the flowering stage, were obtained by hydrodistillation and analyzed by gas chromatography (GC) and gas chromatography/mass spectrometry (GC-MS). Under the optimum extraction and analysis conditions, 44, 38, and 38 constituents (mainly monoterpenes compounds) were identified in T. kotschyanus Boiss. and Hohen, T. eriocalyx (Ronniger) Jalas, and T. daenensis subsp lancifolius (Celak) Jalas which represented 89.9%, 99.7%, and 95.8% of the oils, respectively. The main constituents were thymol (16.4ââ?¬â??42.6%), carvacrol (7.6ââ?¬â??52.3%), and ?-terpinene (3ââ?¬â??11.4%). Antioxidant activity was employed by two complementary test systems, namely, 2,2-diphenyl-1-picrylhydrazyl (DPPH) free-radical scavenging and Ã?Ÿ-carotene/linoleic acid systems. Antioxidant activity of polar subfraction of T. daenensis subsp lancifolius (Celak) Jalas was found to be higher than those of the others in DPPH assay, while nonpolar subfraction of T. eriocalyx (Ronniger) Jalas has most antioxidant activity in Ã?Ÿ-carotene/linoleic acid test (1 9 . 1 Ã?± 0 . 1?Ã?µg/mL and 9 6 . 1 Ã?± 0 . 8% inhibition rate, resp.)....
Background\r\nThe vulnerability of newborn babies' skin creates the potential for a number of skin problems. Despite this, there remains a dearth of good quality evidence to inform practice. Published studies comparing water with a skin-cleansing product have not provided adequate data to inform an adequately powered trial. Nor have they distinguished between babies with and without a predisposition to atopic eczema. We conducted a pilot study as a prequel to designing an optimum trial to investigate whether bathing with a specific cleansing product is superior to bathing with water alone. The aims were to produce baseline data which would inform decisions for the main trial design (i.e. population, primary outcome, sample size calculation) and to optimize the robustness of trial processes within the study setting.\r\nMethods\r\n100 healthy, full term neonates aged <24 hours were randomly assigned to bathing with water and cotton wool (W) or with a cleaning product (CP). A minimum of bathing 3 times per week was advocated. Groups were stratified according to family history of atopic eczema. Transepidermal water loss (TEWL), stratum corneum hydration and skin surface pH were measured within 24 hours of birth and at 4 and 8 weeks post birth. Measurements were taken on the thigh, forearm and abdomen. Women also completed questionnaires and diaries to record bathing practices and medical treatments.\r\nResults\r\nForty nine babies were randomized to cleansing product, 51 to water. The 95% confidence intervals (CI) for the average TEWL measurement at each time point were: whole sample at baseline: 10.8 g/m2/h to 11.7 g/m2/h; CP group 4 weeks: 10.9 g/m2/h to 13.3 g/m2/h; 8 weeks: 11.4 g/m2/h to 12.9 g/m2/h; W group 4 weeks:10.9 g/m2/h to 12.2 g/m2/h; 8 weeks: 11.4 g/m2/h to 12.9 g/m2/h.\r\nConclusion\r\nThis pilot study provided valuable baseline data and important information on trial processes. The decision to proceed with a superiority trial, for example, was inconsistent with our data; therefore a non-inferiority trial is recommended....
We investigated the inhibitory effect of three glycyrrhizin derivatives, such as Glycyrrhizin (compound 1), dipotassium glycyrrhizate (compound 2) and glycyrrhetinic acid (compound 3), on the activity of mammalian pols. Among these derivatives, compound 3 was the strongest inhibitor of mammalian pols a, �Ÿ, ?, and ?, which belong to the B, A, Y, and X families of pols, respectively, whereas compounds 1 and 2 showed moderate inhibition. Among the these derivatives tested, compound 3 displayed strongest suppression of the production of tumor necrosis factor-a (TNF-a) induced by lipopolysaccharide (LPS) in a cell-culture system using mouse macrophages RAW264.7 and peritoneal macrophages derived from mice. Moreover, compound 3 was found to inhibit the action of nuclear factor-?B (NF-?B) in engineered human embryonic kidney (HEK) 293 cells. In addition, compound 3 caused greater reduction of 12-O-tetradecanoylphorbol-13-acetate-(TPA-) induced acute inflammation in mouse ear than compounds 1 and 2. Ixora parviflora with high polyphenol content exhibited antioxidant activity and reducing UVB-induced intracellular reactive oxygen species production. In this study, results of the photoaging screening experiments revealed that IPE at 1000?�µg/mL reduced the activity of bacterial collagenase by 92.7 �± 4.2% and reduced the activity of elastase by 32.6 �± 1.4%. Therefore, we investigated the mechanisms by which IPE exerts its anti-photoaging activity. IPE at 1?�µg/mL led to an increase in type I procollagen expression and increased total collagen synthesis in fibroblasts at 5?�µg/mL. We found that IPE inhibited MMP-1, MMP-3, and MMP-9 expression at doses of 1, 5, and 10?�µg/mL, respectively, in fibroblasts exposed to UV irradiation (40?mJ/cm2). Gelatin zymography assay showed that IPE at 50?�µg/mL inhibited MMP-9 secretion/activity in cultured fibroblasts after UVB exposure. In addition, IPE inhibited the phosphorylation of p38, ERK, and JNK induced by UVB. Furthermore, IPE inhibited the UVB-induced expression of Smad7. In addition, IPE at 1?�µg/mL inhibited NO production and COX-2 expression in UV-exposed fibroblasts. These findings show that IPE exhibits anti-inflammatory and anti-photoaging activities, indicating that IPE could be a potential anti-aging agent....
